CRYSTAL VIOLET STAINING SOLUTION GRAMS
Preparation of solutions Gram Crystal Violet Solution: • Dissolve 20 g of crystal violet in 100 ml of ethanol to make a crystal violet stock solution. Similarly, dissolve 1 g of ammonium oxalate in 100 ml of water to make an oxalate stock solution.
Gram positive bacteria stain violet due to the presence of a thick layer of peptidoglycan in their cell walls, which retains the crystal violet these cells are stained with.
application: Crystal violet is also a mitotic agent and mutagen. It has been used for topical treatment in creams against fungal and bacterial infections. Its been used against some pathogenic fungi such as Candida species and some gram-positive bacteria, e.g. Staphylococcus species.
