Silica and alumina are both polar adsorbents so the more polar components in the mixture to be separated are retained more strongly on the stationary phase and are therefore eluted from the column last. Silica is recommended for most compounds, but as it is slightly acidic, it preferentially retains basic compounds.

Seliwanoff's test is used to distinguish aldoses from ketoses. On treatment with a concentrated Acid, ketoses are dehydrated more rapidly to give furfural derivatives and on condensation with resorcinol give cherry red complex. The test will be answered by fructose, sucrose and other keto containing carbohydrates.

Silica Gel 100-200 Mesh grade of Silica Gel is mostly suitable for gravitational chromatography. This particle size also allows the solvent to flow smoothly without any pressure. Best for a compound that can be easily separated.

Choice of column size is less important than the amount of silica used. However, in practice, most people will choose a column size that allows them to fill column approximately one-third to a half full of silica – not including the solvent reservoir (Fig. 6).

The additive sorbitan monolaurate consists of sorbitol (and its anhydrides) esterified with fatty acids derived from coconut oil. The fluorophore rhodamine B is often used in biological assays. Nonetheless, there are some drawbacks: it can readily form a spirolactam compound, which is nonfluorescent, and therefore may not be the dye of choice for all fluorescence microscopy applications.
